The microbiome and disease: Gut bacteria influence the severity of heart attacks in rats

The microbiome and disease: Gut bacteria influence the severity of heart attacks in rats

Friday, January 13, 2012

New research published online in the FASEB Journal suggests that the types and levels of bacteria in the intestines may be used to predict a person's likelihood of having a heart attack, and that manipulating these organisms may help reduce heart attack risk. This discovery may lead to new diagnostic tests and therapies that physicians use to prevent and treat heart attacks. In addition, this research suggests that probiotics may be able to protect the heart in patients undergoing heart surgery and angioplasty.

"Our discovery is a revolutionary milestone in the prevention and treatment of heart attacks," said John E. Baker, Ph.D., study author from the Division of Cardiothoracic Surgery at the Medical College of Wisconsin in Milwaukee. "The biochemical link between intestinal bacteria, their metabolites, and injury to the heart will reduce the risk of death from a heart attack and, coupled with the use of probiotics, will ultimately be able to improve the overall cardiovascular health of the human population."

To make this discovery, Baker and colleagues conducted experiments involving three groups of rats. The first group was fed a standard diet. The second group was treated orally with the antibiotic vancomycin in the drinking water. The third group was fed a probiotic supplement that contains Lactobacillus plantarum, a bacterium that suppresses the production of leptin.

The group treated with the antibiotic had decreased levels of leptin (a protein hormone that plays a key role in appetite and metabolism), which resulted in smaller heart attacks, and improved recovery of mechanical function as compared to the group fed a standard diet. The antibiotic reduced total bacterial numbers in the intestines and altered the abundance of specific types of bacteria and fungi that live in the gut. Treating these rats with leptin was shown to offset the protection produced by the antibiotic treatment. The third group was fed a probiotic that also altered the numbers and types of bacteria and fungi living in the gut. Like those fed the antibiotic, these rats also had decreased leptin levels, resulting in smaller heart attacks and greater recovery of mechanical function as compared to the first group. Treating these rats with leptin also was shown to offset the protection produced by the probiotic.

"We may not be ready to prescribe yogurt to prevent heart attacks, but this research does gives us a much better understanding of how the microbiome affects our response to injury," said Gerald Weissmann, M.D., Editor-in-Chief of the FASEB Journal. "Just as physicians use cholesterol levels, blood pressure, and overall body composition as measures of heart disease risk, we may soon evaluate our body's susceptibility to disease by looking at the microbes that inhabit the gut."

Space station to move to avoid oncoming junk (AP)

WASHINGTON ? The International Space Station is dodging a softball-sized piece of space junk.

Mission Control told astronauts to fire the station's engines briefly Friday morning to avoid a piece of an old communications satellite.

It will be only the 13th time since 1998 that the station has moved because of debris. Debris travels at such high speed in orbit that it could cause a deadly puncture to the space station.

The last time the station moved was in September. In June, astronauts had to take shelter in the Soyuz escape capsule as debris flew by.

NASA said Thursday that the debris from the private U.S. satellite Iridium was on a path that would have brought it close to the station Friday afternoon, possibly less than a mile.

U.S. slaps sanctions on China state energy trader over Iran (Reuters)

WASHINGTON (Reuters) ? The United States on Thursday imposed sanctions on China's state-run Zhuhai Zhenrong Corp, which it said was Iran's largest supplier of refined petroleum products, as it sought to impress on Beijing and Tehran its resolve to increase economic pressure over Iran's nuclear program.

Secretary of State Hillary Clinton also imposed sanctions on Singapore's Kuo Oil Pte Ltd and FAL Oil Company Ltd, an independent energy trader based in the United Arab Emirates, the State Department said in a notice.

The State Department said the move was part of a broadening international effort to target Iran's energy sector and persuade Tehran to rein in its nuclear ambitions.

"The sanctions announced today are an important step toward that goal, as they target the individual companies that help Iran evade these efforts," the statement said.

The sanction bar all three companies from receiving U.S. export licenses, U.S. Export Import Bank financing or loans over $10 million from U.S. financial institutions, the department said, stressing that the sanctions apply only to the companies and not to their governments or countries.

The U.S. announced the decision after China's rebuff this week of Treasury Secretary Timothy Geithner, who traveled to Beijing to press China on U.S. demands it do more to help curb Iran's oil revenues.

'SHOT ACROSS THE BOW'

Analysts said the U.S. move was largely symbolic, given that Zhenrong was unlikely to have much U.S. business exposure, but that it did send a signal to Beijing and its state-run oil giants such as China National Petroleum Corp (CNPC), China Petroleum and Chemical Corp (Sinopec Corp) and China National Offshore Oil Corp..

These companies have invested billions of dollars in the U.S. energy sector, and are much more exposed to the impact of potential sanctions.

"It's a good shot across the bow and signals the U.S. is serious about vigorous sanctions enforcement. This could be the beginning of a cascade of more sanctions on Chinese companies if China doesn't curtail its Iranian trade," said Mark Dubowitz, executive director of the Foundation for Defense of Democracies, a Washington pressure group that favors stronger sanctions on Iran.

Zhuhai Zhenrong - one of four dominant Chinese state oil traders - brokered the delivery of over $500 million in gasoline to Iran between July 2010 and January 2011 in contravention of U.S. sanctions law, the State Department said.

While the U.S. move targeted Zhenrong for its gasoline sales, the Chinese company has a broader role in Beijing's energy dealings with Iran and has been a major buyer of Iranian oil since at least 1995, typically selling the oil to Sinopec and PetroChina, the country's two dominant refiners.

Zhenrong has been buying about 240,000 barrels per day for several years, representing about 5 percent of China's imports, although sources said last week it would cut crude imports from Iran for a second month in February along with other Chinese oil traders amid a dispute over payments.

In mid-2010, Zhenrong joined Chinese state energy giants in filling a void left by Western oil companies and trading houses that had halted sales of gasoline to Iran because of toughening U.S. sanctions.

Derek Scissors, an expert in the Chinese economy at the Heritage Foundation think tank, said the action against Zhenrong would send a message to other Chinese state oil majors.

"We don't want to be taking action against Sinopec, CNPC and CNOOC. They are huge, and politically powerful," he said.

"But Zhenrong is close enough to them, and won't really do that much harm beyond sending the signal."

TARGETING COMPANIES

The U.S. announcement followed Western moves to tighten the economic noose on Tehran through unilateral sanctions.

President Barack Obama has signed a U.S. law imposing sanctions on financial institutions that deal with Iran's central bank, its main clearinghouse for oil exports, while the European Union is expected soon to agree to a new ban on Iranian oil imports.

Washington has sought to impress on friends and foes that it means business, sending U.S. officials around the world to warn of the dangers of dealing with Iran.

A senior Obama administration official stressed that the purpose of sanctions was to draw Iran back to the negotiating table to discuss curbing its nuclear ambitions, the other half of the 'two-track' U.S. policy of pressure and engagement.

"The theory of the case here is that these two tracks will ultimately converge and Iran will make a decision that it is important to come to the table to try to remove some of these sanctions, to improve their economy," the official told reporters on condition of anonymity.

The other two companies listed by the State Department, both well-known names in the Asian oil trading world, are smaller, private trading firms that typically specialize in shipping bunker fuel or heavy residual products but, like Zhenrong, had also begun doing deals to sell gasoline to Iran.

The State Department said Kuo Oil had provided over $25 million in refined petroleum to Iran between late 2010 and early 2011, while FAL provided over $70 million in refined petroleum to Iran over multiple shipments in late 2010.

In all cases, individual deliveries were worth significantly more than the $1 million threshold under U.S. law and the total value of the transactions was well above the $5 million threshold for sanctionable activities within a 12-month period, the State Department said.

GOP Race Moves South After Romney Wins NH

Former Massachusetts Gov. Mitt Romney won a convincing victory in the New Hampshire primary, the second straight triumph for Romney and one that bolsters his front-runner status to take on President Barack Obama in November. All six Republican contenders head to South Carolina on Wednesday ahead of the next primary. With 95% of precincts reporting, Romney received 40% of the vote in Tuesday's balloting. Texas Rep. Ron Paul received 23% and former Utah Gov. and U.S. Ambassador Jon Huntsman garnered 17%.Romney's sweep of the first two contests for the GOP nomination made history. It was the first time a non-incumbent Republican won both the Iowa caucuses and New Hampshire's first-in-the-nation primary.Former House Speaker Newt Gingrich and former Pennsylvania Sen. Rick Santorum came in with 10% and 9%, respectively, and Texas Gov. Rick Perry had 1%."We didn't compete in New Hampshire. So it doesn't surprise us that our score there was a bit on the low side," Perry said on "Piers Morgan Tonight."With Romney's New Hampshire victory expected, based on polling in recent weeks, the battle for second place and beyond had implications for the South Carolina primary on January 21.Despite the strong showing by Romney, who won nearly every group of voters after his narrow victory last week in the Iowa caucuses, all the other contenders made clear they would continue their campaigns in South Carolina. The Palmetto State will be the first Southern contest of the nomination process and more welcoming to conservatives such as Santorum, Perry and Gingrich, who hails from neighboring Georgia."South Carolina is a winner-take-all state," Perry said. "Winning here, I can promise you, wipes out the caucus victory and New Hampshire. So if Mitt's thinking he's got it in the bag, I think he'll be in for a great surprise in South Carolina when he shows up here."In an interview with CNN before Tuesday's results came in, Gingrich acknowledged South Carolina will be vital to his presidential hopes."We're going to go all out to win South Carolina. We think that's a key state for us," Gingrich said, describing the race there as a contrast between himself -- a "Georgia Reagan conservative" -- and Romney, "a Massachusetts moderate."Romney told CNN Wednesday he does not plan to discount the other candidates and noted he has "a long way to go" to secure the GOP nomination. However, he said, "I really think we're best off focusing on the failures of this president." He said he believes voters want to hear his plans for restoring rising incomes and job growth. In his victory speech in New Hampshire, Romney sounded like the presumptive Republican nominee to take on Obama in November."Tonight we celebrate. Tomorrow we go back to work," Romney told exuberant supporters, calling Obama "a failed president" and asking "the good people of South Carolina to join the good citizens of New Hampshire to make 2012 the year he (Obama) runs out of time."The crowd interrupted with chants of "Mitt" as he outlined a campaign strategy that portrayed Obama as a European-leaning big government advocate while defining his candidacy as a return to American ideals."This president puts his faith in government. We put our faith in the American people," Romney said to cheers.Paul told CNN that he expected to raise more money after a second-place finish, and he then told cheering supporters that their campaign for freedom in America would continue to grow.Referring to Romney, Paul said "he certainly had a clear-cut victory, but we're nibbling at his heels," giving a chuckle as the crowd chanted "President Paul."In response to criticism by rivals that his calls for scrapping the Federal Reserve and bringing home American forces from around the world were dangerous, Paul declared: "We are dangerous, to the status quo."Huntsman told his supporters, "I think we're in the hunt," adding "Hello, South Carolina" to emphasize his third-place finish would keep him in the race.Santorum, who narrowly lost to Romney in Iowa but finished well back in New Hampshire, also said he would head to South Carolina and continued to portray himself as the "true conservative" in the field."With faith in American people we can not only wipe out this deficit and rebuild this economy ... but we can win a huge victory that will rally this country to take on the challenges we have before us," Santorum said.Perry already was in South Carolina in what amounted to a concession of New Hampshire.A record 250,000 voters were expected to turn out in New Hampshire for the GOP primary on an unseasonably warm winter day, Deputy Secretary of State David Scanlan told CNN. With no competitive challenger to Obama on the Democratic side, more "undeclared" voters could weigh in on the Republican race, he said.Exit poll data showed that nearly seven out of 10 Republican voters in the state were very worried about the economy and their personal financial situation. One in four said the deficit was the most important issue. Also, more than three-quarters of respondents said the series of Republican debates was important to their final decision, while less than half said television ads were important. The exit poll data showed that Romney scored a solid victory. Even Christian conservatives and tea party backers -- the heart of Santorum's support last week in Iowa when he lost by just eight votes to Romney -- gave a plurality win to Romney this time.Still, most New Hampshire voters indicated they only made up their minds this week. One of the state's more than 300,000 "undeclared" or independent voters, Linda Underhill, said Tuesday that she decided to support Huntsman. After initially backing Romney, Underhill shifted to Huntsman, calling him smart and likely to take a bipartisan approach."In the past few days, I watched him very closely," Underhill said. "I just feel he is more genuine."Earlier Tuesday, Gingrich argued that a Romney showing in the 30% range, as the most recent polling suggested, could hurt the front-runner even if he won Tuesday's contest."If he can't come close to 50% here, it's very unlikely he can sweep the nomination," Gingrich told reporters in Bedford. "And I think that gives the party time to take a deep breath, look at his record and begin to realize that maybe this isn't the right guy to run against Obama."Gingrich has been pounding at Romney since Iowa, complaining about a massive negative ad campaign against him by allies of the former Massachusetts governor.A Gingrich-allied super PAC has already launched its own anti-Romney barrage in South Carolina, and Gingrich and others have honed in on Romney's years as a financier with Bain Capital, accusing him of getting rich by gutting companies and laying off workers.Romney will have to answer questions about that in conservative South Carolina, Gingrich said Tuesday.Asked about the negative ads from the Gingrich camp, Romney told Boston radio station WRKO on Tuesday that they "will not help" his rival."All I have got to do is keep my head down, keep talking about my message of getting America back to work, my experience in having led two businesses successfully, the Olympics successfully," Romney said.Gingrich wasn't alone in attacking Romney's business record. In South Carolina, Perry told supporters Romney's firm "looted" a photo company in Gaffney and a steel company in Georgetown."I would suggest they are just vultures," Perry said. "They are vultures that are sitting out there on the tree limb waiting for the company to get sick, then they swoop in, they eat the carcass, they leave with that and they leave the skeleton."Asked about those remarks on Wednesday, Romney said the issue has been brought up every time he has run for an office."I understand that President Obama is going to try and put free enterprise on trial," he said. "But, you know, Rick Perry and Newt Gingrich are going to be the witnesses for the prosecution. I'm not worried about that. They can take it as they like, but you saw last night that that approach didn't work very well for either Rick Perry or Newt Gingrich."On Tuesday night, an Obama campaign aide said that despite Romney's victory in New Hampshire, "the premise of his candidacy began to unravel in the last 48 hours," referring to the questions over Romney's business experience.Romney tops most national polling and is ahead in the latest surveys in South Carolina and Florida. However, he came under criticism after a speech Monday to the Nashua Chamber of Commerce, when he said he wanted Americans who were unhappy with their health care coverage to be able to switch insurance companies."I like being able to fire people who provide services to me," he said. "You know, if someone doesn't give me the good service I need, I want to say, 'You know, I'm going to go get someone else to provide that service to me.' "The first seven words of that sentence -- "I like being able to fire people" -- provided an enticing sound bite for opponents to attack."Gov. Romney enjoys firing people. I enjoy creating jobs," Huntsman said at a campaign stop in Concord on Monday.Romney Communications Director Gail Gitcho said opponents are taking Romney's remarks out of context -- a point on which Gingrich and Paul defended him Tuesday. However, the attacks fed the image of Romney that his GOP opponents and Democrats have pushed: That he's a wealthy businessman who can't connect to average Americans.Vice President Joe Biden touched on that theme Tuesday night, telling New Hampshire voters via teleconference that Romney's firing comment was "probably taken a little out of context," but nonetheless reflected Romney's true sentiments."He thinks it's more important for the stockholders and the shareholders and the investors and the venture capital guys to do well than for those employees to be part of the bargain," Biden said. But Romney said Wednesday, "You've got almost 2 million people that have lost their jobs under this president, median income that has dropped 10% over the last few years." Millions are out of work, and some have stopped looking, he said."This is a failed presidency," he said. "People know that. They're going to do their very best to attack whoever the Republican party puts forward. But in the final analysis, they can't defend their record, and it's because of the president's failure that he's going to be replaced, at least in my view."CNN's Paul Steinhauser, Alan Silverleib, Dana Bash, Kevin Bohn, Tom Cohen, Jessica Yellin and Rachel Streitfeld contributed to this report.

'Couch Potato' Pill Prevents Heat Stroke, Study Finds (LiveScience.com)

Scientists have stumbled upon a drug that may prevent heat stroke for people prone to the deadly condition. Ironically, it's the same drug flaunted three years ago as the "couch potato pill" for its ability to build muscle and increase endurance in mice that never break a sweat.

The study appears today (Jan. 8) in the journal Nature Medicine.

As with the 2008 couch potato study, the researchers have only tested the drug, abbreviated as AICAR, in mice. However, the drug was 100-percent effective in preventing death in these mice genetically engineered to be susceptible to heat stroke. Thus, the finding has implications for anyone exposed to heat or with abnormal heat sensitivity, the researchers said.

Heat stroke is caused by prolonged exposure to temperatures over 100 degrees Fahrenheit (37.8 degrees Celsius), and it is common among the elderly and athletes. Many U.S. soldiers in Iraq and Afghanistan suffered from heat stroke brought on by high temperatures and heavy gear.

Some people have a genetic disorder called malignant hyperthermia that places them at high risk for heat stroke, even without the heat. The disorder is associated with a mutation in the RYR1 gene, which causes uncontrolled muscle contractions and increases in body temperature, typically induced by certain drugs such as general anesthesia.

Researchers led by Susan Hamilton at Baylor College of Medicine in Houston studied the effects of the drug AICAR on mice with this RYR1 mutation. When these mice exercise in a hot room, they suffer the hallmarks of malignant hyperthermia and die.

Hamilton's group was inspired by the 2008 AICAR study, published in the journal Cell, which demonstrated how AICAR slowed muscle fatigue and increased muscle endurance. They wondered whether AICAR could control the muscle contractions typical of the RYR1 mutation, but in this case triggered by heat not anesthesia.

Sure enough, AICAR prevented an otherwise sure death in all the mice exposed to heat, even those mice given AICAR just 10 minutes before exercising.

While RYR1 mutations only account for a small percentage of heat stroke cases in the general human population, AICAR might offer broad protection, said Robert Dirksen, a study author at the University of Rochester Medical Center in Rochester, N.Y.

"We think the fundamental process that occurs during heat stroke in individuals with RYR1 mutations is likely to be similar to what happens ... by exposure to even greater temperatures or a longer time" in people without the mutation, Dirksen said. "Our study takes an important first step towards developing a new drug therapy that may be part of the standard treatment regimen for heat stroke."

Although promising, much more research is needed on AICAR and heat stroke, the scientists said, noting that the "couch potato" pill for humans has not come to fruition.

Shaun White's Halfpipe Debuts At Lake Tahoe's Northstar Ski Resort

TRUCKEE, Calif. -- Having an Olympic champion call your local ski resort home has its privileges.

A 22-foot-high high, 500-foot-long halfpipe co-designed by snowboarder Shaun White was opened to the public last week near Truckee at Northstar California, one of Vail Resorts Inc.'s properties formerly called Northstar-at-Tahoe.

The red-haired, two-time Olympic gold medal winner known as the "Flying Tomato" had the superpipe built at the Sierra resort for use in training for upcoming events.

The Verdi, Nev.-based Snow Park Technologies helped design the superpipe, and the resort's crews put in 700 hours in making it.

White claimed gold medals at the Winter Olympics in 2006 and 2010. He signed a deal with Vail in August to represent all of the ski company's resorts in Colorado and California, and announced he was making Northstar his home mountain and primary training resort.
